Abstract

The background and purpose of the simplest Intestinal Nematode worm egg examination is the Native Method using 2% Eosin reagent. The purpose of this study was to determine the difference in the quality of the egg preparation of Soil Transmitted Helminth worms staining purple sweet potato (Ipomea batatas) with Eosin, with the type of experimental analytic research. The results showed that the mean ranks was a reflection of the quality of the coloring of worm eggs by the concentration of purple sweet potato juice and 2% eosin. Concentrations of 1:1, 1:2, 1:4 and 1:5 gave the least good staining quality (mean rank = 10.50) and 1:3 concentration was better with a mean rank of 23.00 and Eosin 2% as control resulted in a mean rank value of 28.00 with a higher value, meaning that the coloring quality of purple sweet potato with a concentration of 1:3 gives the best quality. It can be concluded that variations in the concentration of purple sweet potato juice give a significantly different coloring quality. The staining quality is quite good, namely the concentration of purple sweet potato juice: aquadest (1:3) and the best staining is Eosin 2% (control) with p<0.05.
