Abstract

Tuberculosis (TB) is still a global health problem that requires serious attention. To support government efforts to increase the success rate of TB treatment and diagnosis, accurate early diagnosis is needed for effective TB control. This study aims to compare the results of Mycobacterium tuberculosis examination in saliva and sputum samples using the GeneXpert® Molecular Rapid Test (TCM) and Qualitative Polymerase Chain Reaction (qPCR) methods. This study used an in vitro experimental laboratory design by comparing the results of saliva and sputum sample examinations using the TCM GeneXpert® test kit and RT-qPCR TB Dx in the same patient. MTB in saliva and sputum samples can be detected by both methods. The highest CT value was obtained in sputum samples examined using the TCM method, which was 25.2 and the highest CT value in sputum samples examined using TB Dx, which was 38.85. The results of the study proved that the RT-qPCR method is more sensitive to detecting MTB than the GeneXpert® method, as evidenced by the cut off for the GeneXpert® examination at 25 while the cut off for the RT-qPCR examination was at 38.
