Abstract

Diabetes Mellitus (DM) is a chronic disease with serious complications such as diabetic foot ulcers, which can reduce quality of life and pose a risk of amputation. Ulcer infections are caused by bacteria such as Staphylococcus aureus and Pseudomonas aeruginosa, which thrive in high glucose levels. The biofilm formed by these bacteria makes wounds difficult to heal due to increased resistance to treatment. Although not always directly related to disease severity, biofilm formation can affect psychological conditions and the overall quality of life of patients.This research used a laboratory experimental method with identification techniques including colony morphology, Gram staining, biochemical tests, and confirmation of biofilm formation using Congo Red Agar (CRA) media. The results of this study showed that Staphylococcus aureus formed a strong biofilm, whereas Pseudomonas aeruginosa formed a weak biofilm. It can be concluded from these findings that the bacteria produced biofilms with varying intensities. Keywords: Diabetic ulcer, Staphylococcus aureus, Pseudomonas aeruginosa, biofilm
