Abstract

The nose is an important organ, although it looks small but is quite important. The habit of picking the nose with hands in children can affect nasal health and potentially increase the risk of bacterial infections. This study aims to identify Gram-positive cocci and Gram negative bacilli in the noses of kindergarten children, with a focus on Gram microscopic examination and determination of bacterial types. The research method is descriptive observational, with a cross-sectional approach, conducted on five nasal swab samples from children at Aisyiyah Kindergarten No. 29 Padang. Isolation was carried out using Blood Agar Plate (BAP) and MacConkey Agar (MCA) media, followed by Gram staining and biochemical tests. The results showed Gram-positive cocci bacteria in the form of Staphylococcus aureus and Staphylococcus epidermidis, and Gram-negative bacilli in the form of Escherichia coli and Pseudomonas aeruginosa. This identification indicates that the noses of kindergarten children can be a reservoir of normal flora as well as opportunistic pathogens. The conclusion of this study is that in the noses of kindergarten children, gram-positive cocci were found in 6 specimens and gram-negative bacilli in 4 specimens. There were four types of bacteria found in the noses of kindergarten children, namely Staphylococcus aureus, Staphylococcus Epidermidis, Pseudomonas aeruginosa and Escherichia coli.
