Abstract

Trichosporon asahii is an opportunistic yeast frequently found in patients with diabetes mellitus (DM), particularly those with immune disorders and complications. Conditions such as hyperglycemia, glycosuria, and common DM-related complications create an environment that supports the colonization of this fungus, including in the urinary tract. This study aimed to detect the presence of T. asahii in urine isolates from DM patients with complications using molecular methods including PCR, DNA sequencing, and phylogenetic analysis. This descriptive study with a cross-sectional design used urine samples isolated on Sabouraud Dextrose Agar (SDA) media. Initial identification was performed macroscopically and microscopically, followed by DNA amplification using ITS U1 and U4 primers, DNA sequencing via BLAST GenBank, and phylogenetic tree analysis using MEGA 11. The results showed that out of five isolates, one tested positive for T. asahii. This isolate originated from a female patient with severe DM complications, including nephropathy, neuropathy, and hypertension, and showed 100% similarity to the reference sequence from GenBank (NR_111116.1). Phylogenetic analysis grouped the isolate within the T. asahii cluster with high bootstrap support. The conclusion T. asahii can be found in the urine of DM patients with severe complications.
