Abstract

Diabetes mellitus is a chronic metabolic disease that impairs immune system function, thereby increasing the risk of opportunistic infections, including the fungus Trichosporon asahii. This study aimed to detect the presence of T. asahii in the urine of diabetic patients using culture and Polymerase Chain Reaction (PCR) methods. Urine samples were collected from five diabetic patients with complications at dr. Rasidin Padang General Hospital using the clean catch midstream technique to prevent contamination. Initial identification was performed through culture and observation of macroscopic and microscopic morphology, followed by molecular detection using PCR. The results showed that T. asahii was detected in one sample by PCR, exhibiting higher sensitivity compared to culture. These findings underscore the importance of utilizing both methods simultaneously for early detection of T. asahii infection in diabetic patients. Further research is recommended to incorporate DNA sequencing analysis to strengthen the validity and accuracy of fungal species identification. Thus, the application of molecular methods can enhance the effectiveness of diagnosing and managing opportunistic fungal infections in immunocompromised patients.
