Abstract
Diabetic foot ulcers (DFUs) represent a chronic complication of diabetes melitus (DM) that are highly susceptible to opportunistic infections, including those caused by Trichosporon asahii. This yeast-like basidiomycetes/. fungus is known for its ability to form biofilms, invade deep tissues, and exhibit resistance to several conventional antifungal agents. This study aimed to identify the presence of T. asahii in diabetic foot ulcers using both morphological and molecular approaches. A total of five patients aged 40–70 years with diabetic foot ulcers were examined, consisting of two females and three males, all presenting complications such as peripheral artery disease (PAD), neuropathy, hypertension, nephropathy, and immunodeficiency. Fungal identification was performed through macroscopic culture observation, microscopic examination with Lactophenol Cotton Blue (LPCB) staining, and PCR amplification using ITS1 and ITS4 primers. Sequencing analysis was conducted using MEGA 11 software and the GenBank database. Results indicated that isolate number 4 was positive for T. asahii, characterized by rough-surfaced colonies and microscopic structures including arthroconidia and blastospores. PCR showed a band of approximately 600 bp, and sequencing revealed 99% similarity with T. asahii sequences in GenBank. These findings highlight T. asahii as a significant opportunistic pathogen in diabetic foot infections, and support the use of ITS-based molecular identification as a reliable diagnostic tool in non-Candida fungal infections.
