Abstract

Breast milk (ASI) is the main source of nutrition for infants, offering a complete and ideal combination of nutrients to support early growth and development. Breast milk (ASI) is one source of LAB that maintains the balance of the digestive tract microflora. The use of LAB in health as a probiotic can increase immunity, help overcome digestive disorders, and potentially prevent the growth of cancer cells in the digestive tract. In addition, breast milk can increase the baby's immunity and reduce the risk of anemia in mothers. This study aims to isolate and characterize LAB from breast milk. The research was conducted at the Biomedical Laboratory of Pioneer University of Indonesia from June 2024 to June 2025. This research method is an experiment with a non-probability sampling technique. Data were analyzed using descriptive tests. The results showed the presence of a clear zone around the colony on MRS agar media which indicates the ability of LAB to dissolve CaCO₃. The conclusion of this study is that lactic acid bacteria have been successfully isolated from breast milk. Gram staining showed the presence of bacilli and Gram-positive cocci. The catalase test showed a negative result, indicated by the absence of gas bubbles after the addition of H₂O₂. The suggestion in this research is to conduct biochemical and molecular tests to gain a deeper understanding of lactic acid bacteria. Keywords: Breast Milk (ASI), Lactic Acid Bacteria, Isolation
