Abstract

Chronic Suppurative Otitis Media (CSOM) is a middle ear infection often caused by Pseudomonas aeruginosa, a Gram-negative bacterium known for its ability to form biofilms and resist various antibiotics. Virgin Coconut Oil (VCO) contains active compounds such as lauricacid that possess antibacterial properties and hold potential asan alternative therapy.This study aimedtoidentifythe presenceof P.aeruginosa intheblood of CSOM – induce dratmodels followingtreatment with VCOusingtheconventional PCRmethod. Thestudywas conducted experimentally with five groups of rats, consisting of a positive control group, a negative control group, and three treatment groups receiving VCO at concentrations of 40%, 80%, and 100%. Blood samples were analyzed using conventional PCR with 16S rRNA primers to detect P. aeruginosa DNA. The results showed that the treatment groups, particularly the group receiving100%VCO,exhibited reduce dorun detectable DNA bandintensity compared to the positive control group,which showed specific P. aeruginosa DNAbands. Itcan beconcluded that VCO effective lyreducesthepresence of P.aeruginosa in the blood of CSOM ratmodels. Further studies using larger sample sizes and quantitative approaches are recommended to strengthen these findings.
