Abstract

Candida Albicans is an opportunistic fungus that causes oral candidiasis, particularly in individuals with weakened immune systems such as patients with type 2 Diabetes Mellitus (DM). This study aimed to detect the presence of the Internal Transcribed Spacer (ITS) gene of C. albicans in the oral mucosa of type 2 DM patients. The research was conducted using an analytical observational design with a cross-sectional method on 10 oral mucosa swab samples from type 2 DM patients. Initial identification was performed macroscopically and microscopically, followed by a germ tube test and detection of the ITS gene using PCR with ITS1 and ITS4 primers.The results showed one positive isolate of C. albicans characterized by convex, shiny white colonies, yeast cells (blastospora), budding cells, and pseudohyphae. The germ tube test was also positive with hyphal outgrowth formation. PCR confirmed the presence of the ITS gene of C. albicans. PCR is proven to be more effective and accurate than conventional methods in detecting C. albicans. Keyword : Candida Albicans, mucosa, Polymerase Chain Reaction (PCR).
