Abstract

Senduduk leaf (Melastoma malabathricum L) is one of the traditional medicinal plants in West Sumatra which is rich in phytochemicals such as phenyls, flavonoids and saponins. This study aims to determine the phenolic content and antioxidant activity of the acid hydrolyzed fraction of senduduk leaf extract. Hydrolysis was carried out in an acidic environment using HCl on senduduk leaf extract which had its non-polar part removed. The total phenolic content of the hydrolyzed fraction was determined using the Folin Ciocalteu method, while the antioxidant activity was examined using the DPPH radical method. Based on the results of the study, the total phenolic content of the non-hydrolyzed fraction and the 2N, 4N, 6N hydrolyzed fraction were 4.87644%w/w; 5.22516%b/b; 5.25827%b/b; and 6.3233%w/b. Antioxidant activity was expressed by IC50 values for fraction A of 91.45 ppm (strong), fraction B 79.72 ppm (strong), fraction C 78.04 ppm (strong), and fraction D 57.64 ppm (strong). It can be concluded that hydrolysis with 2N, 4N, 6N HCl increased the total genolic content and antioxidant activity. Keywords: Melastoma malabatricum L, antioxidant, phenolic, fraction, extract
