Abstract
Staining in microscopic examination of Soil-Transmitted Helminths (STH) eggs aims to clarify the morphological shape for easy observation and identification. Eosin 2% is a commonly used dye, but has several disadvantages such as potential irritation, toxic effects, and limited availability in some areas. This study aims to determine whether the boiled water of paper flowers (Bougainvillea glabra choisy) can be used as an alternative dye to Eosin 2% in the examination of STH worm eggs, and to assess the effectiveness of staining based on various concentrations of paper flower solution. The method used was a laboratory experiment with five dye treatments (25%, 50%, 75%, 100% concentration, and 2% Eosin as control), and five repetitions on STH positive fecal samples. The results showed that concentrations of 75% and 100% provided the best staining quality, with clarity of egg shape and color contrast close to the results of 2% Eosin staining. Based on these results, paper flower cooking water has the potential to be an effective, safe, and environmentally friendly alternative natural dye for microscopic examination of STH eggs.
