Abstract

ABSTRACT Diabetic foot ulcer is a chronic complication of diabetes mellitus that often leads to lower extremity amputation. Its healing process is frequently aggravated by infections caused by both Gram-positive and Gram-negative bacteria. This study aimed to identify and characterize the bacterial pathogens responsible for diabetic foot ulcer infections. An experimental study was conducted at the Microbiology Laboratory of Universitas Perintis Indonesia in June 2025. Samples were collected using the swab method from the wound surface of patients and cultured on Blood Agar Plate (BAP), MacConkey Agar (MCA), and Mannitol Salt Agar (MSA). The isolates were observed macroscopically, Gram-stained, and tested using catalase, coagulase, various biochemical assays (TSIA, Citrate, SIM, MR-VP), and carbohydrate fermentation tests (glucose, lactose, maltose, sucrose). The results revealed three bacterial pathogens: Staphylococcus aureus, Pseudomonas aeruginosa, and Klebsiella pneumoniae. S. aureus showed positive catalase and coagulase reactions, P. aeruginosa did not ferment any tested sugars, while K. pneumoniae fermented all tested sugars. These findings highlight the importance of microbiological examination in guiding appropriate therapy for diabetic foot ulcer infections.
