Abstract
Tuberculosis (TB) remains a major public health concern in Indonesia. One of the frequent complications is secondary respiratory infection caused by other bacteria, such as Haemophilus influenzae, Escherichia coli, Pseudomonas aeruginosa, and Enterobacter cloacae. This study aimed to detect co-infecting pneumonia bacteria in TB patients using the Multiplex Real-Time PCR method with the Crown Lab® Pneumoplex 3.0 kit. Sputum samples were collected from clinically confirmed TB patients. DNA extraction was followed by amplification of target genes using a CFX96 Real-Time PCR instrument. The results showed simultaneous detection of multiple bacteria with varied Ct values. Haemophilus influenzae was the most frequently detected pathogen, followed by E. coli and P. aeruginosa. The Multiplex RT-PCR method proved to be sensitive and specific, enabling rapid and efficient detection of co-infection in TB patients. Therefore, this molecular approach supports more accurate diagnosis and targeted therapy. Keyword : Tuberkulosis, ko-infeksi, pneumonia, multiplex PCR, Crown Lab Pneumoplex 3.0.
