Abstract

Chronic suppurative otitis media (CSOM) is a chronic inflammation of the middle ear with perforation of the tympanic membrane and purulent discharge that can cause hearing loss. This study aims to assess the effect of Virgin Coconut Oil (VCO) on the presence of Pseudomonas aeruginosa DNA in CSOM model mice and the potential of VCO as a natural antibacterial. The research method used an in vivo post-test only control group experimental design in male mice divided into five groups: negative control, positive control, and VCO treatment of 40%, 80%, and 100%. Detection of P. aeruginosa DNA was carried out using 16S PCR to show a significant decrease in DNA band intensity after VCO administration. The results showed that VCO concentrations of 80% and 100% were able to reduce the presence of P. aeruginosa DNA, while at 40% it was still detected after VCO administration. This indicates that VCO has the potential as a natural therapeutic agent for CSOM infections. The conclusion of this study is that 16S rRNA PCR is a reliable method for evaluating the effectiveness of natural treatments, and the use of VCO shows promise as a natural-based supportive therapy in the treatment of CSOM. Suggestions are directed towards further research using sequencing methods for more in-depth bacterial identification.
